Dopamine (DA) activity was measured in 25 unmedicated patients with panic disorder using the prolactin (PRL) release test. The mean PRL peak value in 13 of 25 patients was elevated as compared to that of 16 normal control subjects, both of whom had been drug free for 4 days. Diagnosis and classification of prostate carcinomas has a large impact on patients' management. Cytogenetic studies have shown that the majority of prostate carcinomas have recurrent genetic lesions, most of which are consistent with changes that may occur during prostatic cancer progression. We have used a combination of fluorescent in situ hybridization (FISH) and comparative genomic hybridization (CGH) to identify new genetic lesions in an array of prostate carcinomas. Chromosomes 4, 8, 10, 13, 16, 17, 18, 21, and X were included in the panel of FISH probes. Three DNA samples of different prostate carcinomas were tested using these probes. When a novel genetic lesion was detected in the tumors, it was further analyzed using CGH. This combination of techniques has led to the identification of six new genetic lesions in prostate cancer, four of which are mutations. These four are the first known missense mutations in oncogenes in prostate cancer. One of the mutations involves the protooncogene KRAS in a prostate carcinoma, whereas the other three involve tumor suppressor genes. The importance of these genetic lesions is currently under investigation.
